What is Semax?

Semax is a synthetic peptide based on a fragment of the hormone ACTH (4–10), studied in brain and neuropeptide research.

ACTH — adrenocorticotropic hormone — is a 39-amino-acid hormone with a well-known role in the stress-response system. Semax takes only residues 4 to 10 of that sequence, a seven-amino-acid fragment, and adds a short Pro-Gly-Pro tail at one end. The choice of fragment is the interesting part: the region responsible for ACTH's hormonal activity sits elsewhere in the molecule, so the fragment used here carries the neuromodulatory characteristics of the parent hormone without its endocrine ones.

The Pro-Gly-Pro extension is a stability modification. Short peptides are cleared quickly by enzymes in biological media, and that tripeptide tail — a motif found in naturally occurring regulatory peptides — substantially slows degradation. It's a recurring design pattern in this field: take a fragment that carries the activity you want, then bolt on something that stops it being broken down before it can be studied.
